چکیده مقاله:
Abstract Background and Purpose: Candida parapsilosis is a common cause of candidemia in children and onco-hematologic patients as well as in patients with septic arthritis, peritonitis, vaginitis, and nail and skin infections. Here, we evaluated intra- and inter-species beta tubulin DNA sequence variation within the C. parapsilosis complex with a view to establishing its utility in the identification and phylogenetic analysis of the species. Materials and Methods: The novel primers successfully amplified the target for all 23 strains included in the study. Results and Conclusion: Multiple alignment of 623–629 bp-long nucleotide (nt) sequences reflecting the beta tubulin gene indicated an inter-species divergence ranging from 0 to 68 nt in C. parapsilosis, C. orthopsilosis and C. metapsilosis with a mean similarity of 84.7% among the species. Meanwhile, intra-species differences of 0–20 and 0–6 nt were found within the strains of C. parapsilosis and C. orthopsilosis, respectively. The phylogenetic tree topology was characterized by a clade made up by C. parapsilosis and C. orthopsilosis, while C. metapsilosis formed a related but separate lineage. Our data provide the basis for further discoveries of the relationship between the species belonging to the C. parapsilosis complex and the value of beta tubulin DNA sequence data in the identification and taxonomy of C. parapsilosis and other pathogenic yeasts.

Candida parapsilosis Characterization in an Outbreak Setting
Candida parapsilosis is an important non-albicans species which infects hospitalized patients. No studies have correlated outbreak infections of C. parapsilosis with multiple virulence factors. We used DNA fingerprinting to determine genetic variability among isolates from a C. parapsilosis outbreak and from our clinical database. We compared phenotypic markers of pathogenesis, including adhere...
متن کاملCharacterization of the Candida parapsilosis complex in East China: species distribution differs among cities.
Candida parapsilosis, which was previously considered to be a complex of three genetically distinct groups, has emerged as a significant agent of nosocomial infections. Recently, this complex was separated into three species: C. parapsilosis sensu stricto, C. orthopsilosis and C. metapsilosis. In China, data pertaining to these fungi are limited. In this study, we examined 57 isolates of member...
متن کاملThree distinct genotypes within Candida parapsilosis from clinical sources.
Three genetically distinct groups of Candida parapsilosis were detected among clinical isolates. These were distinguishable on the basis of isoenzyme profiles and DNA sequences of internally transcribed spacer (ITS) sequences flanking the 5.8S RNA gene. In an investigation of 45 strains, including 32 clinical isolates from Texas, C. parapsilosis group I composed the majority of the common clini...

Background and Purpose: Candida parapsilosis is a predominant species found in nosocomial infection, particularly in hospitalized patients. The molecular epidemiology of the clinical strains of this species has not been well studied. The present study was performed with the aim of investigating the microsatellite genotyping of Candida parapsilosis among the Iranian clinical isolates. Materials...
متن کاملCandida parapsilosis complex induces local inflammatory cytokines in immunocompetent mice.
Despite the increasing incidence of the Candida parapsilosis complex in the clinical setting and high mortality rates associated with disseminated infection, the host-fungus interactions regarding Candida parapsilosis sensu stricto and the closely related species C. orthopsilosis and C. metapsilosis remains blurred. In this study, we analyzed inflammatory cytokines levels and histopathology as ...
متن کاملLooking into the virulence of Candida parapsilosis
Editorial In many diagnostic microbiology laboratories the species identification process starting from cultured colonies has recently undergone some significant changes. Where not long ago car-bon/nitrogen source utilization tests in the form of colorful tubes or well-based devices down to chip card size dominated the field, we now find a purely biophysical instrument, a MALDI-ToF mass spectro...
